Ag-Tetrahydrocannabinol from Whole Blood Using the Zymark RapidTrace with Confirmation and Quantitation by GC-EI-MS
نویسنده
چکیده
A sensitive, reliable, and automated solid-phase extraction (SPE) method was developed for the simultaneous extraction, confirmation, and quantitation of Ag-tetrahydrocannabinol (THC) and 11-nor-9-carboxy-A%tetrahydrocannabinol (THCCOOH) from whole blood. The extraction was performed on the Zymark RapidTrace SPF Workstation with a reduced solvent volume SPE copolymer cartridge. Quantitative analysis was completed on a benchtop gas chromatograph-mass spectrometer using electron ionization mode with selected ion monitoring of three ions for each analyte. The limits of quantitation for THC and THCCOOH were 2 ng/m/and 1 ng/rnL, respectively. The limits of detection for THC and THCCOOH were 1.6 ng/mL and 0.8 ng/mL, respectively. Extensive method validation is presented including extraction recoveries, within-run precision, day-to-day precision, linearity, and carryover. This procedure is used routinely in the laboratory on blood samples screened positive for cannabinoids.

Cannabinoids present in the blood samples were quantified using solid-phase extraction ...
متن کاملQuantitation of benzodiazepines in whole blood by electron impact-gas chromatography-mass spectrometry.
Benzodiazepines are frequently encountered in forensic toxicology. A literature search was conducted to find a simple method using electron impact-gas chromatography-mass spectrometry (EI-GC-MS) to examine whole blood specimens for the most commonly encountered benzodiazepines in the United States. A recently published method was identified in the literature search and used as a starting point ...
متن کاملConfirmation of THC in Oral Fluids Using High-Resolution 2-D GC/MS
Oral fluids are being used as an alternative matrix to urine for drug testing. Oral fluid is considered to be less invasive and much more difficult to adulterate than urine samples. Samples of oral fluid are typically screened by an ELISA immunoassay method. Those found to be positive must be further analyzed with a mass spectrometry confirmation technique. The confirmation technique must be ab...
متن کاملAcute nitrobenzene poisoning with severe associated methemoglobinemia: identification in whole blood by GC-FID and GC-MS.
A rare fatal case of self-poisoning with nitrobenzene following oral ingestion is reported. On presentation to the hospital, severe methemoglobinemia (70%) was observed in an 82-year-old male who had ingested 250 mL of an unknown substance in the previous 24 h. Methylene blue and exchange transfusion were the therapeutic methods applied in the treatment of the methemoglobinemia. Forty-eight hou...
